| 内容記述 | 京都府立大学農場ホルスタイン種乳牛より得られた牛乳より, Armstrongらの分別法に従いβ-ラクトグロブリン, α-ラクトアルブミン画分を分別し, それぞれをSephadex G-100でのゲル滬過及びDEAE-Sephadex A-50でのイオン交換クロマトグラフィーにより精製した。Armstrongらの方法により得られたβ-ラクトグロブリン及びα-ラクトアルブミン画分はCellogel電気泳動の結果から, 多くの他種蛋白質を含んでいることが示されたが, ゲル滬過によりかなり精製され, さらにイオン交換クロマトグラフィーによりほとんど純粋にされうることが電気泳動及びアミノ酸分析の結果から明らかとなった。又β-ラクトグロブリン異型体はpH6.3でのDEAE-Sephadex A-50クロマトグラフィーで完全に遺伝的変異体A及びB型に分離されうる。α-ラクトアルブミンはDEAE-Sephadex A-50クロマトグラフィーにおいて非常に複雑な挙動を示したが, これは透析あるいはクロマトグラフィーの間, 分子の一部に不可逆的な立体構造の変動か自己会合が生じ荷電が変化した結果であると考察した。 | |||||

| 内容記述 | A study has been made of the column chromatography of bovine β-lactoglobulin and α-lactalbumin. The β-lactoglobulin and α-lactalbumin fractions fractionated by the Method I-a and II-a of Armstrong et al. were purified by gel filtration on Sephadex G-100. Further purification was effected on DEAE-Sephadex A-50 ion-exchange chromatography at pH 6.3. β-Lactoglobulin could be separated into two protein peaks by the DEAE-Sephadex column chromatography. Amino acid analyses and Cellogel electrophoreses indicated that these protein peaks correspond to A and B variants of β-lactoglobulin, respectively. On the othe hand, α-lactalbumin exhibited the complex behaviour on the DEAE-Sephadex chromatography. This is believed to be due to the irreversible conformational changes or the self-association which may result in the changes of charge. | |||||
