| 内容記述 | The procedure of a modified squash method applicable to the young leaf, petal as well as root tip is described briefly as follows : (1) Macerate the material in 1.2N. HCl for 6-25 mins. (2) Rinse in water about 30 secs. (3) Immerse in chomo-acetic mixture (CrO_3 1g., glacial acetic acid 3cc., distilled water 100cc.) for 10-120 secs. (4) Rinse in water about 30 secs. (5) Immerse in ammonia alcohol (30% alcohol 100cc., ammonia 5-6 drops) for 10-30 secs. (6) Rinse in water about 30 secs. (7) Place the material on a slide mounted with a drop of aceto-carmine (45% acetic acid, 1.5g carmine) under a cover glass. (8) Apply firm pressure to squash. (9) Pass the slide quickly over a spirit flame 6-7 times. (10) Press slightly on the cover glass. This method is satisfactory for the cells with small chromosomes which tend to become clumped and difficult to spread out by a usual aceto-carmine squash method after the use of HCl for maceration. Chromo-acetic mixture is better than formalin or alcoholic fixatives. Acetic-lacmoid can be used in place of aceto-carmine in this procedure. Ammonia alcohol makes the figures clearer. The preparation sealed with wax are kept in good condition for the observation for about three months. | |||||
