Natural science and living science}, month = {Nov}, note = {びわ果実アセトンパウダーよりポリフェノールオキシダーゼを抽出し, 30∿90%硫安分画にて酵素を部分精製した。本酵素の等電点は, 焦点電気泳動よりpH 3.5,また分子量はSephadex G-100のゲル濾過より42,000と推定した。本酵素はο-ジヒドロキシフェノールに対して広い基質特異性を示し, その至適pHは6.8(ピロカテコール, D-カテキン)と5.9(4-メチルカテコール)であった。熱に対しては, 本酵素は60℃, 30分間処理ではほとんど失活せず, 80℃, 30分間処理でほぼ失活した。また, pH 4∿6の弱酸性域で最も安定であった。, A polyphenol oxidase was extracted from acetone powder of loquat with 0.1M Naphosphate buffer, pH 6.5 and partially purified by 30∿90% ammonium sulfate fractionation. Isoelectric focusing and Sephadex G-100gel filtration of the preparation indicated that the enzyme had an isoelectric point of 3.5 and a molecular weight of 42,000. The enzyme showed wide substrate specificity toward ο-dihydroxyphenols. Optimum pHs were 6.8 when pyrocatechol and D-catechin were used as substrate and 5.9 in case of 4-methylcatechol as substrate. The enzyme was stable at 60℃ for 30 min but it lost more than 95% of the activity after 30min at 80℃.
