Agriculture","bibliographic_titleLang":"en"}]}]},"item_3_description_10":{"attribute_name":"抄録(日)","attribute_value_mlt":[{"subitem_description":"通常,植物のcDNAライブラリー構築には数十∿数百グラムの材料を必要とし,器官レベルまでの構築が限界であった。本研究では,PCR法を用いイネ種子一粒を出発材料にして組織,細胞レベルのcDNAライブラリー構築を目指した。その結果,登熟イネ種子の三分の一粒相当から得たアリューロン層,デンプン性胚乳に対応するcDNAライブラリーを構築することが出来た。胚乳ライブラリーからは1.3kbpのグルテリンcDNAクローン,0.6kbpの全長プロラミンcDNAクローンの存在を確認した。同様に,アリューロン層cDNAライブラリーからは数種のアリューロン層特異的クローンを得ることができた。これらのことより,この方法を用い組織,細胞レベルでの遺伝子発現機構が厳密に調査可能であることが判った。","subitem_description_language":"ja","subitem_description_type":"Other"}]},"item_3_description_11":{"attribute_name":"抄録(英)","attribute_value_mlt":[{"subitem_description":"We constructed a cDNA library from a single developing rice grain. RNA was obtained from a sheet of manually peeled aleurone layer from one third of a single rice grain. Single stranded cNDAs were amplified using the PCR technique after oligo (dG) tails were introduced on the 3'-ends of the single stranded cDNAs obtained directly from the mRNAs.
